Human Cancer Exome (Onconome) Panels

We are pleased to announce our new MYbaits Onconome KL v1 series of cancer exome target enrichment kits. Both of these validated NGS capture kits target thousands of exons in hundreds of cancer-related genes described in two major studies:

  • Kandoth et al. 2013. "Mutational landscape and significance across 12 major cancer types". Nature 502:333-339, doi:10.1038/nature12634
  • Lawrence et al. 2014. "Discovery and saturation analysis of cancer genes across 21 tumour types". Nature 505:495-501, doi: 10.1038/nature12912

Each baitset was developed through multiple design iterations in order to improve coverage uniformity and filter out repetitive genomic loci. Many of these loci are implicated in a variety of human phenotypes, not just cancer. Therefore the panel is an excellent, cost-effective choice for exploratory targeted exome sequencing in several research settings, such as human ancient DNA studies.

Two baitset kits are available: the KLv1.0 Panel which targets ~1.1Mb from 283 genes, and the expanded KLv1.5 Panel which targets an additional ~400Mbp of exons from a total of 290 genes.

Onconome KL v1.0 Panel

This panel targets over 5,200 exonic intervals (6,500+ annotated exons) from 283 cancer-related genes (see Table 3 below). Total target length is 1.1 Mbp of the human genome. When US$15 in sequencing is applied, >88% of these positions routinely yield coverage close to the overall mean observed (within 0.2-3.0× of mean unique coverage), and >94% of targeted bases are covered to a unique coverage depth of at least 10×, with >85% at 20× and >79% at 30× (see Fig. 1).

Onconome KL v1.5 Panel

This panel expands on KLv1.0 by targeting over 2,000 additional unique exonic intervals, including those from seven additional cancer-related genes, bringing the total target length to nearly 1.5Mbp. (Additional targeted genes are FOXA2, HRAS, MALAT1, RPS15, SOX17, SRSF2, TCEB1; see full list of genes covered in Table 3).
